2-Pyrrolidineacetic Acid and Pyrrolizidine Alkaloids from Melampodium divaricatum.

چکیده

The atypical, non-toxic pyrrolizidine alkaloid tussilagin and its isomers have been found in only a few plants of the Asteraceae (Roeder et al., 1981,1984; Passreiter et al., 1992; Roeder et al., 1993; Sener and Etgun, 1996; Wernery et al., 1997; Passreiter, 1998). Their possible biosynthetic precursor pyrrolidine-2-acetic acid and its methylester were isolated and identi"ed together with the pyrrolizidine alkaloids tussilagin, isotussilagin and their C-1 epimers neo-tussilagin and neo-isotussilagin from Arnica species and Tussilago farfara (Passreiter, 1992). Later on both types of compounds were also found in Neurolaena lobata (Passreiter, 1998).

Melampodium divaricatum is a medicinal plant, which occurs in Central America. In a recent paper we reported the occurrence of acylated 2-hydroxy thymol glycosides as main constituents in this plant. This paper deals with the isolation of two new 2,5-dihydroxythymol ester derivatives. The formerly reported sesquiterpene lactone mikanokryptin was not found in our plant material.

Pyrrolizidine Alkaloids from Heliotropium

Investigation of the pyrrolizidine alkaloids of Heliotropium arbainense resulted in the isolation and identification of heliotrine, europine and 7-angelylheliotrine. From H. ovalifolium three alkaloids were isolated: supinine, 7-angelylheliotridine and an alkaloid which was shown to be of the heliotridine monoester type with a dehydrated viridifloric acid.

Spithioneines A and B (1 and 2), two new bohemamine-type pyrrolizidine alkaloids possessing an unusual ergothioneine moiety, were isolated from a marine-derived Streptomyces spinoverrucosus. Their structures were elucidated by spectroscopic analysis, CD spectra, and chemical degradation and synthesis. Compounds 1 and 2 are rare natural products that incorporate the amino acid ergothioneine.
